چکیده

The relationship between molecules having granulocyte colony-stimulating activity (G-CSA), erythroid burst-promoting activity (E-BPA), and activity promoting increase in the number of granulocytic progenitors in liquid culture ( GPA) was explored in conditioned medium from human leukocytes (HLCM) and human placenta (HPCM). As tested on human hemopoietic progenitors in culture. G-CSA eluted from Sephadex G100 as a single peak with apparent molecular weight of 25,000, separating partially from EBPA and GPA, which both had an apparent molecular weight of 45.000. All three activities eluted together from hydroxyapatite at low molarity phosphate. Their charge properties were also similar and all three electrofocused in flat gel beds in the pH range near 5.4. On both hydroxyapa-

Macromolecules stimulating human granulocytic colony-forming cells, precursors of these cells, and primitive erythroid progenitors: some apparent nonidentities.

The relationship between molecules having granulocyte colony-stimulating activity (G-CSA), erythroid burst-promoting activity (E-BPA), and activity promoting increase in the number of granulocytic progenitors in liquid culture (delta GPA) was explored in conditioned medium from human leukocytes (HLCM) and human placenta (HPCM). As tested on human hemopoietic progenitors in culture, G-CSA eluted...

Effect of flt3 ligand on the ex vivo expansion of human CD34+ hematopoietic progenitor cells.

A ligand for the tyrosine kinase receptor flt3/flk-2, referred to here as flt3 ligand (flt3L), was recently cloned. The effect of flt3L on purified human CD34+ progenitor cells was examined. flt3 receptor (flt3R) was detected on the surface of human bone marrow cells that were enriched for CD34 expression. The effects of flt3L and the c-kit ligand Steel factor (SLF) on hematopoietic progenitors...

Differential and synergistic effects of human granulocyte-macrophage colony-stimulating factor and human granulocyte colony-stimulating factor on hematopoiesis in human long-term marrow cultures.

The ability of granulocyte-macrophage colony-stimulating factor (GM-CSF) and G-CSF to influence hematopoiesis in long-term cultures (LTC) of human marrow was studied by cocultivating light density normal human marrow cells with human marrow fibroblast feeders engineered by retroviral infection to constitutively produce one of these growth factors. Feeders producing stable levels of 4 ng/mL GM-C...

Granulocytic and Monocytic Differentiation of CD34hi Cells Is Associated With Distinct Changes in the Expression of the PU.l-Regulated Molecules, CD64 and Macrophage Colony-Stimulating Factor Receptor

The present study investigated the possibility that macrophage colony-stimulating factor (M-CSF) responsiveness of hematopoietic progenitor cells is regulated at the level of receptor expression and that M-CSF receptor (M-CSFR) may be used as an early marker of monocyte lineage commitment. Immunofluorescence measurements with an antiM-CSFR antibody showed that 44% * 5% of CD34h' cells expressed...

Effects of human interleukin-3 on granulocytic colony-forming cells in human bone marrow.

Human multilineage colony-stimulating factor (multi-CSF)/interleukin-3 (IL-3) induces colony formation from CFU-GEMM, BFU-E, and CFU-Eo when applied to in vitro cultures of highly enriched hematopoietic progenitor cells. No granulocytic colonies are formed in response to IL-3. However, with appropriate assays, we demonstrate that IL-3 increases the size of G-CSF-induced granulocytic colonies; t...
